Sequencing Sample Submission Guidelines

 

The following concentrations and volumes are recommended guidelines. 

Typically, we request more volume than we need - if you need to send less, just let us know.

 

Sample Concentration Volume
PCR Product <500bp*** ~40ng/µl 5-10µl
PCR Product >500bp*** ~60ng/µl 5-10µl
Plasmid DNA single or 96-well* ~400ng/µl 5-10µl
Primer**** 8pmol/µl 5-10µl
Bacterial Samples-Low copy 109-1010 cells/ml 1-2mls
Bacterial Samples-High copy 108-109 cells/ml 1-2mls
Bacterial Samples-96-well** 108-109 cells/ml 100µl
Stab or Streak cultures 1 stab or plate -
BAC clone ~300ng/rxn -

 

*DNA should be resuspended in molecular grade water, elution buffer or as a dried down pellet. Do Not Store In TE.  Lower concentration of DNA may be used in some cases.

 

**Bacterial Samples sent in 96-well format should be stored as glycerol stocks.

 

***Please do not dilute your PCR product.  Typically, PCR is within the concentration range necessary for sequencing.

 

****We prefer the primers to be sent separately, not pre-mixed with the template.  Thanks.
